You figure out how fast you walk and spray to figure out how many gallons of solution you are applying per acre (or per 1000 square feet). Then you know what concentration to mix.
You could start by spraying out a water-only backpack load in a square section of the barn lot to figure out your overall carrier rate (gallons per 1000 sq ft or acre). The odds are, with a backpack sprayer, that your rate is really high, like 50 gallons per acre or higher. Once you have adjusted your nozzle spray pattern you will want to keep it the same, if possible.
Of course you should always follow the label but if you are just preventing weeds from germinating in a barnlot, crop injury is not a concern, so if you happen to accidentally overapply due to inaccurate calibration of your walking speed while spraying, your odds of having trouble are low.
