If air isn't leaking past the bead at 45 psi give it a day or two, the pressure could slowly get the bead to fully seat if it's lubed. I had that problem with a couple tractor tires once. Each got pumped up to 35 psi and left up on a jack. 24 hours later they were seated. Tire lube should work best, it won't dry like soap.
