The oil test kits I get from Cat have a small hose that you can dip in the fill hole to pull a sample without draining the oil. I think you want to run it first to mix things up.

In your situation, I would be more concerned with getting the engine hot regularly to burn off the water. If you are only warming it up in the spring, it might make sense to change the oil, or consider warming it up every few months.
