For tissue I'd recommend taking both old and new to compare.

If you have the money, a sap test will give you a more nuanced look. For example you can have adequate iron in your tissue sample but it will be in the form or iron oxide (rust) and not usable to the plant. A sap test only shows plant available nutrients. Kind of like the difference between doing a nutrient test on blood vs your entire arm.
