It is unusual to have sound for a while then it stops.
Audi and wifi are two areas which updating to W10 has caused problems but it is usually driver related.
Your best option will be try roll back the driver and see if any other 'basic' driver perhaps from Microsoft is available already on the machine, via device manager.

Also try to get exact part numbers of the device and google for driver packs preferably from the maker.
This is a potentially risky area to be downloading stuff - avoid any kind of programs offering to scan and search for you. 

It does sound like the kind of thing will mysteriously fix itself after a couple of reboots.
You might also try some kind of alternative speakers if possible.
It seems a bit unclear if the actual jack is somehow unresponsive rather than the speakers themselves.
