According to my "slide rule" and calculations.... a 10 inch, single wall pipe on .5% (.005) grade can move about 32,000 gallons of water per hour.  I'm not sure if your scenario of running 2/3 full requires a simple reduction of 1/3 of the 32,000.   I would think it would.  If so, you should be around 20,000 gallons of water per hour.

As to the second part of your question.... personally, I don't think the 12 inches of submergence would cause much a reduction of flow.  But I certainly could be wrong.
