You can't kill it if you try. The stalks get old and tough, but if you have new stalks, pick away. The stalks get smaller as the plant runs out of energy, so if the stalks are decent size, you know you'll have plenty next year. Frost can cause the poison in the leaves to move down to the stalk, don't eat any rhubarb after a frost.
