Run a few cal loads at different flow rates.
Do one as fast as you think you can go with the combine
Then do one at your typical speed and yield.
Do one at 1/2 your typical seed/yield.

In your monitor, you can look at the grain flow rate. You want to have the flow as constant as possible while you are running the cal loads. You might have to speed up/down to keep the grain flow steady at your target -- for example, try to cut the entire cal load at 2000 bu/hr flow rate (or whatever)
Keep you cal loads the same weight.....ie, make them all 6000# or 10000#.

I had a new midrange combine this year and had to do several cal loads at different rates to get it accurate for different yields/speeds.
