I have banded lots of 100 lb calves, never gave tetanus vaccine, and never had a problem.  My experience!

Banded a fair number of 400 and up and always use the vaccine at banding and so far no problems.  But have heard numerous horror stories from those who haven't used the vaccine.

I think the difference is that on the bigger calves the band has to "cut" through more tissue and greater opportunity to create an anaerobic site where tetanus can grow and flourish......

That all said, technically your vet is right because generally it takes some time for the immune system to respond to a new antigen (which in this case is the vaccine)....
