| Changing the resolution does not affect the layout if you choose a resolution in the same aspect ratio. What can also change the layout is changing the DPI (dots per inch) settings between 90 and 120 DPI, but even then it wouldn't affect web pages, mostly Windows dialog boxes.
But you're right, past a certain resolution, you won't see the benefit in a traditional desktop application, it will benefit mostly video playback, games and some graphic applications. But these are already optimized to take full advantage of your monitors native resolution regardless of the virtual resolution set on the desktop, so set up once for all the screen resolution that works best for you on the desktop, at the distance you are watching your monitor, and enjoy your multiple monitors. You won't have to zoom in or adjust the resolution ever to read your emails or IMs as they pop up on the second monitor.
Multiple monitors have better support on Windows 8, but it's ... Windows 8! :)
